Mechanism of action
It acts by inhibition of synthesis of progesterone and thromboxane synthesis through inhibition of both COX-1 and COX-2. This causes reduction in pain, inflammation and swelling.
It follows similar action as that of lornoxicam. Paracetamol is a weak inhibitor for prostaglandin synthesis. It decreases the PG concentration but does not produce action against inflammation in arthritis.
After oral administration it is rapidly absorbed in GI tract. About 90-100% of the drug is absorbed.
The plasma protein binding of the drug is about 99%. The drug is metabolized by cyp 2C9.
The drug is excreted in unchanged form in urine. Nearly about 2/3 of the drug is excreted by liver and 1/3 of the drug by kidney in active form.
It is rapidly absorbed from the GI tract with the volume of distribution about 50L.
It is metabolized in liver into its toxic and non-toxic products.
About 85-90% of the drug is excreted in urine.
