Product Description
Prazoto DSR Capsules are the combination of pantoprazole and domperidone in sustain release form.
Pantoprazole
It is a proton pump inhibitor which inhibits the secretion of gastric acid. It belongs to the same class as that of omeprazole.
Domperidone
It is a specific blocker of dopamine receptor. It is used to relieve nausea, vomiting, to increase the transit of food through the stomach.
Indication
- Short term treatment for erosion of stomach
- Ulceration of esophagus caused by GERD
- Maintenance therapy for long term use after initial response
- In treatment with H. pylori infections
- Nausea and vomiting
- Gastroparesis
- Domperidone is useful in parkison’s disease
- Domperidone may be used in functional dyspepsia in both adults and children.
- Symptomatic relief of erosive or ulcerative GERD
- Reduces heart burn symptoms
- Day and night term heart burns associated with GERD
- Relief in duodenal ulcer
Mechanism of action
Domperidone
It increases the peristaltic stimulation. It facilitates the gastric emptying and decreases the small bowel transit time by increasing the esophageal and gastric peristalsis. The antiemetic property is related to blocking of dopamine activity.
Pantoprazole
It suppresses the final step in gastric acid production by forming a covalent bond to the sites of secretory surface. The effect of pantoprazole is dose related.

Absorption
After oral administration it is well absorbed in the body.
Distribution
The drug is metabolized by the liver with Cmax about 2.5mcg/ml. the peak concentration of the drug is about 2.5hrs. The apparent volume of distribution is 11.0-23.6 L.
Excretion
About 18% of the drug is excreted in feces through biliary excretion.

About 91%-93% of the drug is bound to plasma protein.

About 31-66% of the drug is excreted through urinary and fecal secretion.
Contraindications
- The drug overdose cannot be removed
- Limb splay
- Lateral position
- Absence of ear reflex
- Tremor
- Hypersensitivity to any of the ingredient
Precautions
- Atrophic gastritis
- Acute intestinal nephritis
- Long treatment with acid suppressants can cause cyanocobalamin deficiency
- Clostridium difficile associated diarrhea
- Increased risk of oateoporosis
